Chondroitin sulfate (CS) is a member of glycosaminoglycan (GAG) family in the extracellular matrix of connective tissue and blood vessel walls. GAG has been shown to play a role in cell proliferation, migration and wound healing. In this study, we investigated the protective effects of CS on rat urinary bladder against cisplatin-induced cytotoxicity. We treated the isolated bladder with CS before cisplatin administration and examined the protective effects of CS on the biochemical and pathological changes in the rat bladder, and the relationships between the degree of protection and the dosage of CS. Rats were divided into 6 groups (n=5). Animals in CS groups received 0.5-1.0mg/ml of CS solution orally for 3 weeks. The control group was treated with 0.9% saline solution, the normal group with no treatment, and the Cis group with administration of cisplatin (20mg/kg, i.p.). The CS+cisplatin group received 0.5mg/ml of CS solution orally for 3 weeks before cisplatin administration, and the 1.0mg/ml group received 1.0mg/ml of CS solution. The urinary volume and weight, and weight of bladder tissue, were lower in the CS groups than in the Cis group. The change of protein content in the bladder tissue was higher in the CS groups than in the Cis group. The MTT assay revealed that cell viability in the bladder tissue was higher in the CS groups than in the Cis group. The level of 3-nitrotyrosine in the bladder tissue was lower in the CS groups than in the Cis group. In conclusion, CS has the potential to protect rat urinary bladder against cisplatin-induced damage. CS may be useful as a candidate protective agent against nephrotoxicity and nephrotic syndrome induced by cisplatin.Pages
